Nowadays, all my experiments are in silico, I don't need reagents, I don't need cells or animals to test experiments. My experiments happen 100% on computers. The data though comes from living beings, humans or animals. But in the past, I used to work with the regular "wet lab" experiments and had some contact with animal drug testing. Chemical/biological compounds must be tested before coming to us as final customers, which is us. How does that happen? Why do we still need to use animals?
I will use the example of the internship experience that I had during my undergrad studies. The focus was to cure leishmaniasis. I skin disease caused by a protozoan and the treatment is very difficult since they are eukaryotic organisms, like us! So their cell is very similar to ours, different from bacteria. So I was working in the preclinical part of drug testing. We usually started with experiments in vitro. First just the culture of protozoa. If something doesn't work in that stage you can throw it in the trash immediately since you find the protozoa directly in contact with bovine serum and other compounds that keep them alive while the test continues. But if something succeeds in killing them, we bring it to the next stage in vitro which is using mammal cells. We add to the culture immune mouse cells in the culture so the protozoan infects it and stays inside of it. If something that worked in the previous stage didn't work in this stage, it means that probably the compound couldn't enter the mammal cell. So that means we can't continue with this chemical.
What about now? Is it safe already to go to human consumption? Still no. We aren't just a blob of cells in a culture medium. We are complex organisms and chemical compounds in our bodies can have different directions. Is the compound going to be delivered where we want in the skin? Is the compound going to cause a side effect? That's why we need to continue testing a complex organism, but the safer way usually is through animals. At that time we used mice. Many mice died during these experiments for sure. We used to infect their legs with leishmaniasis and test the chemicals that passed through the in vitro testing.
However, we had rules on how to kill these animals! There is something called bioethics and for every experiment with living beings, we used to have permission from the bioethics department in the University. During that time the more humanized way to kill animals was with overdose of inhaled anesthetics and CO2. So this way we could see how the chemical could work in a complex organism before starting the human trials. That was 13 years ago when I had this experience.
Nowadays, some other possibilities can work in simpler tests, like topical cosmetics. People can grow entire organs in laboratories using stem cells. Not a whole organism. So for example I know someone that grows human skins in petri plates. To test a shampoo, a topical cream is perfect. You don't need to use living beings again since we are talking about a chemical with a local effect. But still,l it is challenging for an oral or intravenous compound that needs to travel through a whole organism. So I respect cosmetics businesses that abandoned animal testing. Animal testing is a necessary evil until we don't find another solution like that.
I told once the experience of my brother with pre-clinical research and he became vegan after that. But for me, we still rely a lot on animal experiments. It is sad to see another animal dying for tests, but we can reduce the guilt by using ethical methods that reduce the pain during their final moments. Human organs that were grown in the lab can be an alternative to reduce the killing but still, we are far away from totally eliminating this type of experiment. And we are saving many human lives by animal testing first.
Hoje em dia, todos os meus experimentos são in silico, não preciso de reagentes, não preciso de células ou animais para testar experimentos. Meus experimentos acontecem 100% em computadores. Os dados, porém, vêm de seres vivos, humanos ou animais. Mas, no passado, eu costumava trabalhar com experimentos regulares de "laboratório úmido" e tive algum contato com testes de drogas em animais. Compostos químicos/biológicos devem ser testados antes de chegarem até nós como clientes finais, que somos nós. Como isso acontece? Por que ainda precisamos usar animais?
Vou usar o exemplo da experiência de estágio que tive durante meus estudos de graduação. O foco era curar a leishmaniose. Uma doença de pele causada por um protozoário e o tratamento é muito difícil, pois são organismos eucarióticos, como nós! Então, a célula deles é muito parecida com a nossa, diferente das bactérias. Então, eu estava trabalhando na parte pré-clínica dos testes de drogas. Geralmente começávamos com experimentos in vitro. Primeiro, apenas a cultura de protozoários. Se algo não funcionar nesse estágio, você pode jogá-lo no lixo imediatamente, pois encontra o protozoário diretamente em contato com soro bovino e outros compostos que os mantêm vivos enquanto o teste continua. Mas se algo consegue matá-los, levamos para o próximo estágio in vitro, que é usar células de mamíferos. Adicionamos à cultura células imunes de camundongo na cultura para que o protozoário o infecte e permaneça dentro dele. Se algo que funcionou no estágio anterior não funcionou neste estágio, significa que provavelmente o composto não conseguiu entrar na célula do mamífero. Então isso significa que não podemos continuar com este produto químico.
E agora? Já é seguro ir para o consumo humano? Ainda não. Não somos apenas uma massa de células em um meio de cultura. Somos organismos complexos e compostos químicos em nossos corpos podem ter direções diferentes. O composto será entregue onde queremos na pele? O composto causará um efeito colateral? É por isso que precisamos continuar testando em um organismo complexo, mas a maneira mais segura geralmente é por meio de animais. Naquela época, usávamos camundongos. Muitos camundongos morriam durante esses experimentos, com certeza. Costumávamos infectar suas pernas com leishmaniose e testar os produtos químicos que passavam pelo teste in vitro.
No entanto, tínhamos regras sobre como matar esses animais! Existe algo chamado bioética e para cada experimento com seres vivos, costumávamos ter permissão do departamento de bioética da Universidade. Naquela época, a maneira mais humanizada de matar animais era com overdose de anestésicos inalatórios e CO2. Então, dessa forma, podíamos ver como o produto químico poderia funcionar em um organismo complexo antes de começar os testes em humanos. Isso foi há 13 anos, quando tive essa experiência.
Hoje em dia, algumas outras possibilidades podem funcionar em testes mais simples, como cosméticos tópicos. As pessoas podem cultivar órgãos inteiros em laboratórios usando células-tronco. Não um organismo inteiro. Então, por exemplo, conheço alguém que cultiva peles humanas em placas de Petri. Para testar um xampu, um creme tópico é perfeito. Você não precisa usar seres vivos novamente, pois estamos falando de um produto químico com efeito local. Mas ainda assim, é desafiador para um composto oral ou intravenoso que precisa viajar por um organismo inteiro. Então, respeito as empresas de cosméticos que abandonaram os testes em animais. Os testes em animais são um mal necessário até que não encontremos outra solução como essa.
Contei a experiência do meu irmão com a pesquisa pré-clínica e ele se tornou vegano depois disso. Mas para mim, ainda dependemos muito de experimentos com animais. É triste ver outro animal morrendo para testes, mas podemos reduzir a culpa usando métodos éticos que reduzem a dor durante seus momentos finais. Órgãos humanos que foram cultivados em laboratório podem ser uma alternativa para reduzir a matança, mas ainda estamos longe de eliminar totalmente esse tipo de experimento. E estamos salvando muitas vidas humanas testando animais primeiro.
